Представители рода Pseudoxanthomonas, ранее преимущественно выделяемые из разных загрязненных почв, в настоящее время описываются как новый оппортунистический патоген человека.</p></sec><sec><title>Цель исследования</title><p>Цель исследования. Биологическая характеристика, филогенетический анализ и определение белкового спектра для идентификации штамма P. kaohsiungensis, потенциально нового и редкого патогена человека.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Объектом исследования стал штамм P. kaohsiungensis IMB-1, выделенный от ребенка, который проходил лечение в стационаре городского уровня. Выделение и культивирование штамма проводили на кровяном агаре. Изучали морфо-биохимические свойства. Для идентификации использовали рибосомную филогению; дополнительно получали белковые спектры с помощью MALDI-TOF масс-спектрометрии.</p></sec><sec><title>Результаты</title><p>Результаты. Со среды обогащения из образца ликвора были изолированы мелкие серые колонии. Морфология клеток – грамотрицательные палочки, оксидазо- и каталазо-положительные. Идентификация с помощью тест-системы NEFERMtest 24 не дала однозначный результат. Культура была чувствительна при повышенном режиме дозирования к триметоприму-сульфаметоксазолу. Белковый спектр с низкой степенью оценки и сходимостью показал первичную идентификацию как Stenotrophomonas maltophilia. По результатам секвенирования по Сэнгеру была собрана последовательность гена 16S рРНК. Первичная идентификация по базе данных NCBI выявила максимальную гомологию с типовым штаммом P. kaohsiungensis J36, которая составила 99,8 %. Белковые спектры, полученные с помощью MALDI-TOF масс-спектрометрии, были обработаны с использованием специализированного программного обеспечения Microbe Analysis и сформирован референсный белковый профиль для идентификации штаммов вида P. kaohsiungensis.</p></sec><sec><title>Заключение</title><p>Заключение. Трудности в бактериологической идентификации клинических значимых новых штаммов бактерий, отсутствие референсных белковых спектров в базах данных подтверждают золотой стандарт молекулярных методов секвенирования и рибосомной филогении для точной идентификации и характеристики патогена. Полученные результаты подчеркивают острую необходимость в усилении геномного мониторинга и обновлении баз данных для быстрой идентификации условных патогенов.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Background</title><p>Background. Although previously isolated from contaminated soils, new opportunistic human pathogens belonging to the genus Pseudoxanthomonas have been described. Analysis of the biological properties of a P. kaohsiungensis strain isolated from the blood indicated that it is a potential human pathogen in immunocompromised patients and not merely a bacterium that produces biosurfactants in the environment.</p></sec><sec><title>The aim</title><p>The aim. To conduct biological characterization, phylogenetic analysis, and protein spectroscopy to identify the P. kaohsiungensis strain, a potential new and rare human pathogen.</p></sec><sec><title>Materials and Methods</title><p>Materials and Methods. The P. kaohsiungensis strain IMB-1 was isolated from a child undergoing treatment in a city hospital. The strain was isolated and cultured on blood agar. Morphological and biochemical properties were studied. Ribosomal phylogeny was used for identification; additionally, protein spectra were obtained using MALDI-TOF mass spectrometry.</p></sec><sec><title>Results</title><p>Results. Small gray colonies were isolated from an enrichment medium from cerebrospinal fluid sample. The cell morphology was Gram-negative rods, oxidase- and catalase-positive. Identification using the NEFERMtest system was inconclusive. The strain was susceptible to increased doses of trimethoprim-sulfamethoxazole. Protein spectrum with a low score and convergence indicated a primary identification as Stenotrophomonas maltophilia. A 1470-bp 16S rRNA gene sequence was assembled using Sanger sequencing. Primary identification using the NCBI database revealed 99.8 % homology to the type strain of the species P. kaohsiungensis. Protein spectra obtained using MALDI-TOF mass spectrometry were processed, and a reference protein profile was generated for identifying P. kaohsiungensis strains.</p></sec><sec><title>Conclusion</title><p>Conclusion. Difficulties in bacteriological identification of clinically significant new bacterial strains and the lack of reference protein spectra in databases highlight the importance of genomic tools for accurate pathogen identification and characterization and confirm the gold standard of molecular sequencing and ribosomal phylogeny.
